RAW 264.7°ú ÀÌ°ÍÀÌ ºÐÈ­ÇÑ ÆÄ°ñ¼¼Æ÷¾ç ¼¼Æ÷¿¡¼­ ÆÄ°ñ¼¼Æ÷ ºÐÈ­°ü·ÃÀ¯ÀüÀÚÀÇ ÀüüÀûÀÎ À¯ÀüÀÚ ¹ßÇöÀ» Á¶»çÇϱâ À§ÇØ cDNA array ¹æ¹ýÀ» »ç¿ëÇÏ¿´´Ù. 1176 cDNA spot grid°¡ ÀÖ´Â Mouse Atlas cDNA array °á°ú¸¦ È®ÀÎÇϱâ À§ÇÏ¿© ¿ªÀü»ç È¿¼Ò ÁßÇÕ¹ÝÀÀ °Ë»ç¸¦ ½ÃÇàÇÏ¿´´Ù. cDNA array °á°ú 6°³ÀÇ À¯ÀüÀÚ°¡ 2.5% ÀÌ»ó ¹ßÇöÀÌ Áõ°¡ÇÏ¿´À¸¸ç(PKC beta¥±, POMC, PTEN µî), 16°³ÀÇ À¯ÀüÀÚ°¡ 2.5%ÀÌ»ó ¹ßÇöÀÌ °¨¼ÒÇÏ¿´´Ù(Osteopontin, Cyclin D1, Cathepsin C, PTMA µî), PKC beta¥±À¯ÀüÀÚÀÇ ¿ªÀü»ç-È¿¼Ò ÁßÇÕ ¹ÝÀÀ °Ë»ç °á°ú ÀÌ À¯ÀüÀÚ¸¦ È®ÀÎÇÒ ¼ö ÀÖ¾ú´Ù. To examine the global gene expression of osteoclastogenesis-related genes in RAW 264.7 and its differentiated OCLs through the use of Atlas Mouse cDNA Array 2.1 membranes printed with 1176 well-characterized mouse genes involved in biology. Both samples were screened in parallel using cDNA expression arrays. The array results were additionally validated using RT-PCR. The results of cDNA arrays showed that 6 genes were up-regulated >2.5-fold (PKC beta 1l. POMC, PEN, etc) and 16 genes were down-regulated )2.5-fold (Osteopontin, Cvclin Cathepsin C, PTMA, etc) in both samples at the mRNA level. RT-PCR analysis of PKC beta II of these differentially expressed genes gave result consistent with cDNA array findings. The result of osteoclastogenesis showed that the PKC beta II gene was overexpressed in OCLs compared with RAW264.7 cell line. Osteoclastogenesis-related genes are differentially expressed in RAW264.7 cell line and its differentiated OCLs. its gene overexpression correlates with osteoclast differentiation in RAW264.7 cell line.
